The separation and purification of black rice anthocyanins and oxidation resistance

 

Abstract: On black rice anthocyanins purification and study the antioxidant activity of the different parts. First of all, using the method of solvent extraction will crude extracts from black rice anthocyanins in accordance with the size of polarity into petroleum ether soluble parts and soluble in ethyl acetate parts and n-butyl alcohol soluble and water soluble parts, and each part removal of 2, 2 - diphenyl - 1 - bitter hydrazine free radical (DPPH ·) and hydroxyl free radical activity are analyzed,  and determine the antioxidant capacity of the strongest component part is water soluble. Second with macroporous resin column chromatography method for water soluble part of the segment, C1 and C2, C3, C4 and C5 five components, the removal of components and the DPPH · free radicals and the activity of hydroxyl radicals was analyzed, and determine the antioxidant capacity of components from big to small in the order: C2 > C3 > C4 > C1 > C5.

    In recent years, active oxygen and free radical research become the frontier and hot issue in modern life science, as a natural antioxidant, anthocyanins compounds are widely used in many fields such as medicine, food, cosmetics, electrochemical. Black rice anthocyanins belong to flavonoids, polyphenols, is made up of anthocyanins with one or more glucose, rhamnose, galactose, xylose and arabinose formed by the combination of glycosidic by glycosidic bond, mainly including mallow element, geranium - 3, 5 - two glycosidase, cornflower element - 3 - glucoside and cornflower - 3, 5 - two glycosidase (its structure as shown in figure 1), usually several anthocyanins coexist. Research shows that black rice anthocyanins have strong anti-oxidant, anti-inflammatory, anti-tumor, allergy, fall hematic fat, such as a variety of functions, and its biological activity because of the B ring is different with the chemical structure of certain differences, this requires monomer composition of black rice anthocyanins in clear and further study of its physiological functions such as antioxidant activity.

    At present, a great deal of research about the black rice anthocyanins on the extraction methods of black rice anthocyanins, less study on the separation and purification methods. Research of black rice anthocyanins purification method and oxidation resistance of different structure of black rice anthocyanins, helps to reveal the black rice anthocyanins antioxidant activity and its structure, the relationship between the provide theoretical basis for the rational use of black rice anthocyanins. Black rice resources, is characteristic of black rice anthocyanins pigment substances, so studying the characteristics of black rice anthocyanins, development and comprehensive utilization of black rice has important guiding significance.
